Only one instance of Marlow will spawn at once, and a new one won't spawn until you kill her or reload the save. Upon killing Marlow 10 times, she will transform into her DangerMario form. Once you defeat DangerMario, Marlow will no longer spawn naturally.

Marlow is rather dangerous, and I wouldn't reccomend engaging her in combat until you have at least prot 4 diamond gear.
